Plan to read and draft the background introduction for Activity Based Protein Profiling (ABPP), which is the proteomics method to profiling proteins based on their functions. The method was established by Dr. Cravatt from Scripps.
Research
Zinc Sulfate and Methanol work for protein precipitation from cell culture media. But only using diethyl ether was not enough to get good recovery rate. Today, I will try different liquid liquid extraction condition to get good recovery rate.
I really like the way you break up your daily goals into writing and research goals... I think I need to do that too. Very specific reading / research goals in the afternoons/evenings would help me produce more writing in the morning. Sounds like you're making steady progress!

ReplyDeleteI'm excited to learn how your liquid liquid extraction goes. I've just started to use liquid liquid extraction for pulling atrazine and metabolites out of spent culture media from a bioreactor.
ReplyDeleteHow are you able to keep on track with experiments after the writing group? The first two days, I didn't really have the enthusiasm to go work in lab after writing.
Yes. I still try to carry out some experiments after 4-hour writing. It is pretty tough. But when I put my goals on the blog, it makes me eager to finish them.
DeleteI tried dimethyl ether from the old paper, not working well. Yesterday I tried hexane: dimethyl ehter 1:1 and hexane: MTBE 1:1. Hexane: Dimethyl ether works better but the recovery rate is still low. So I need to find some other conditions today
research as you write- I think it's a super smart plan!!!!!!! If only I had had time to do that while I was reading old documents in dusty archives
